Abstract :The anti-tumor and other physiological activities of an extract of the bark of Rhizophora stylosa Griff obtained from Iriomotejima Island were investigated in vitro and in vivo. The water-soluble fraction of a methanol extract obtained from air-dried bark was separated by liquid-liquid partition using ethyl acetate. The ethyl acetate fraction was then separated by Sephadex LH-20 column chromatography to obtain methanol:water and acetone:water fractions. We then investigated the cytotoxicity of the four fractions in RAW264.7 cells. Survival rates at the lowest doses of all samples were very high. The effects of three different doses of these fractions on the NO production in LPS-treated RAW264.7 cells were investigated. The cells produced 140 μmol/L of NO in the presence of 40 μg/ml of the ethyl acetate fraction, which was about 1.6-fold more than that produced by control cells incubated with PBS. The ethyl acetate fraction was eluted once again through Sephadex LH-20 using ethanol/methanol, which generated 10 fractions (Fr.1 through 10). Murine peritoneal macrophages produced more NO when incubated with these fractions than with standard catechin. The ethyl acetate fraction (3 mg/day/mouse) was also administered intraperitoneally to ICR mice bearing sarcoma-180 tumors every other day for about 10 weeks. Although the survival of the mice did not significantly increase, tumor metastasis and volume tended to be suppressed. The ethyl acetate fraction of Rhizophora stylosa Griff increased NO production in RAW264.7 cells and in murine peritoneal macrophages, and suppressed the metastasis and proliferation of sarcoma-180 tumor cells. These findings suggest that this fraction contains useful immunoregulators.
西表島産ヤエヤマヒルギ(Rhizophora stylosa Griff)の樹皮抽出画分について,抗腫瘍活性等を in vitro およびin vivo で検討し,以下の成績を得た.風乾樹皮からメタノール抽出物を得,これの水可溶画分を酢酸エチルで液・液分配して酢酸エチル画分を得た.さらに酢酸エチル画分から Sephadex LH-20 カラムクロマトグラフィーによってメタノール:水溶出画分とアセトン:水溶出画分を得た.これら4試料についてRAW264.7株細胞を用いて細胞毒性試験を実施した結果,いずれの試料も0.39mg/ml の最低用量において高い生存率を示した.この結果を踏まえて4試料についてそれぞれ3濃度(400~20μg/ml)を設定し,LPS刺激下でのRAW264.7株細胞のNO産生量に及ぼす影響を調べた結果,酢酸エチル画分の40μg/mlの濃度において対照(PBS)の約1.6倍相当,140μmol/LのNO産生が見られた.酢酸エチル画分を再度 Sephadex LH-20 カラムクロマトグラフィーに付し,エタノール・メタノール溶出により10画分(Fr.1~10)に細分画し,それぞれの25μg/ml暴露下におけるマウス腹腔内マクロファージのNO産生に及ぼす影響を調べた結果,Fr.2~Fr.9において標準カテキンよりも高いNO産生がみられた.また,Sarcoma-180 担癌ICRマウスの腹腔内に酢酸エチル画分(3mg/mouse/day)を約10週間にわたって隔日投与したところ,マウスの生存率では有意な延命効果を得るには至らなかったが,腫瘍の転移および腫瘍容積において抑制傾向がみられた.これらの結果から,酢酸エチル画分にはRAW264.7株細胞あるいはマウス腹腔内マクロファージのNO産生促進作用や Sarcoma-180 腫瘍細胞の転移ならびに増殖の抑制作用があると考えられ,有用な免疫調整物質が含まれている可能性が示唆された.
